  • Prepared in accordance to the following procedure¹:

FORMULATION:

  • Solution #1
    • Dibasic potassium phosphate………………34 g
    • Deionized water……………………………1000 mL
  • Solution #2
      • Magnesium chloride…………………………81.1 g
      • Deionized water……………………………1000 mL

PREPARATION:

  • Sterilize both solutions by steam sterilization
  • Store refrigerated
  • To prepare a working solution add 1.25 mL of Solution #1 and 5 mL of solution #2 to deionized water and make up to 1 L
  • Adjust pH to 7.2 ± 0.2
  • Dispense into appropriate containers
  • Sterilize either by autoclaving for 15 minutes at 15 lbs pressure and 121 °C (glass test tubes or glass bottles) or by gamma irradiation (plastic flip-top bottles)

QUALITY CONTROL:

  • If final volume is less than 90 mL
    • Prepare pour plates of samples using 1 mL of buffer and plate count agar
    • Incubate for 72 hours at 35 °C
  • If final volume is = 90 ml
    • Filter sample through a 0.22 μ filter
    • Aseptically remove the filter and place on surface of a plate count agar plate
    • Incubate at 35 °C for 72 hours
